The Nittany Lions edged out the Dukes 64-62 in the last meeting in December 2014, which was a home contest for Penn State.
Penn State leads the series 27-19. Duquesne leads the series 13-8 at Pittsburgh.
Last season, Penn State finished with an 18-16 record, including 14-2 in non-conference play.
Penn State returns two starters in sophomore guard Shep Garner (9.2 ppg) and senior forward Brandon Taylor (9.1 ppg).
